Note that the above animation shows fully duplex DNA. In the region marked "duplex" above, this is a valid representation. In the region marked "melted" it is not.

Strong evidence has been found for major groove contacts from position -5 through -11.

However, in the region marked melted, identification of contacts indicates that either 1) the enzyme wraps around to the "back side" of the DNA or 2) the duplex view is incorrect and the DNA is melted. In this case all recognition contacts lie along the template strand in a melted context. Our evidence strongly supports this latter view.
